【背景知识】

胰岛素样生长因子 (IGF)-1 和 -II 发挥作用在调节多种细胞类型的增殖、分化和特定功能中起关键作用 (1-3)。IGF-l 与 somatomedin C(Sm-C) (4) 相同，分子量为 7649 道尔顿 (5)。它的主要调节剂是生长激素 (WHi) 和食物。

与许多其他激素不同，IGF 以高亲和力与特异性结合蛋白 (IGFBP) 结合。目前已知有七种不同的结合蛋白 (7,8,22)，它们要么以相似的亲和力结合 IGF-I 和 IGF-II，要么偏好 IGF-II (9,10)。测定 IGF-I 的一个主要问题是测定中对 IGFBP 的干扰。在未处理的血清样品中直接测定 (11) 会导致不正确的结果，因为由于 IGF-I/IGFBP-3 复合物的解离速度极慢，因此在测定孵育期间只有部分 IGF-可用于测量。根据样品中 IGF-/ 与 IGFBP 的比率，会发生干扰。
